Tagging with xxxDDDDK may be done at the N-terminus, N-terminus preceded by a methionine residue, C-terminus, and in internal positions of the target protein. The small size of the epitope tag and its high hydrophilicity tend to decrease the possibility of interference with protein expression, proteolytic maturation, antigenicity and function. Application

An ELISA plate is coated with 50 μL of a DDDDK tag conjugated BSA at concentration of 10 μg/mL. The peptide conjugated carrier protein is coated and detected with anti-DDDDK tag antibody at concentration ranged from 1.95 to 125 ng/mL.
Application

DDDDK tag antibody detects DDDDK tag at cytoplasm and nucleus by immunofluorescent analysis. Samples: HeLa cells mock (left) and transfected with ZNF365–fusion protein (right) were fixed in 4% paraformaldehyde at RT for 15 min. Green: DDDDK tag stained by DDDDK tag antibody diluted at 1:1000. Blue: Hoechst 33342 staining. Scale bar = 10 μm.
